Formation of methyl sterols in brain cholesterol biosynthesis. Sterol formation in vitro in actively myelinating rat brain.

چکیده
A study was made of the methyl sterols involved in sterol biosynthesis in developing rat brain. In addition to the determination of the endogenous sterols present in 15-day-old rat brain, experiments were conducted to establish which of these sterols could be labeled by a minced tissue preparation in the presence of [Z-W]mevalonic acid. The turnover of [14C]lanostero1, [4-3H]squalene oxide, and [14C]squalene by minced preparations was also shown. In the experiments involving incubation of labeled mevalonic acid with minced tissue preparations, the diunsaturated methyl sterols had a larger degree of labeling than the monounsaturated methyl sterols. Labeled squalene was also shown to be present in the mevalonate incubations. Under these in vitro conditions cholesterol was not found to be significantly labeled. Endogenous sterols of two types were examined, those occurring as steryl esters and those that were unesterified. A comparison of the distribution of sterols in these two fractions indicated a relatively small percentage of cholesterol (54.3%) in the steryl ester fraction. Based on gas chromatographic retention data and gas chromatography-mass spectrometry, the following sterols were identified: lanosterol, 4,4-dimethyl-Sot-cholesta-8,24-dien-3fi-01, 4,4-dimethyl-Scr-cholest-8-en-3/3-01, 4cr-methyl-Sty-cholest-g-en3fi-01, 4a-methyl-S~-cholesta-8,24-dien-3~-ol, 4cu-methylSa-cholesta-7,24-dien-3fi-01, 4a-methyl-Sa-cholest-7-en-3/301, Sty-cholesta-7,24-dien-30-01, desmosterol, and cholesterol.

متن کاملPurification of a soluble rat liver protein that stimulates microsomal 4-methyl sterol oxidase activity.
A soluble rat liver protein that stimulates microsomal methyl sterol oxidase activity has been isolated and purified to homogeneity by salt fractionation, differential heat inactivation, two calcium phosphate gel association, and Sephadex filtration. The protein, as isolated, may be dissociated with detergent into subunits with a molecular weight of approximately 10,300, as determined electroph...
متن کاملBrain sterols in the AY-9944 rat model of atypical absence seizures.
PURPOSE The AY-9944 (AY)-treated rat is a reproducible and clinically relevant animal model of atypical absence seizures. AY inhibits cholesterol synthesis, but the relation between brain sterol levels and the spontaneously recurrent absence seizures has not been determined. METHODS Long-Evans hooded rats were treated every 6 days from postnatal day (P)2 to P20 with AY (7.5 mg/kg, s.c.) or sa...
متن کاملDietary intake of plant sterols stably increases plant sterol levels in the murine brain.
Plant sterols such as sitosterol and campesterol are frequently administered as cholesterol-lowering supplements in food. Recently, it has been shown in mice that, in contrast to the structurally related cholesterol, circulating plant sterols can enter the brain. We questioned whether the accumulation of plant sterols in murine brain is reversible. After being fed a plant sterol ester-enriched ...
متن کاملAberrant pathways in the late stages of cholesterol biosynthesis in the rat. Origin and metabolic fate of unsaturated sterols relevant to the Smith-Lemli-Opitz syndrome.
Minor aberrant pathways of cholesterol biosynthesis normally produce only trace levels of abnormal sterol metabolites but may assume major importance when an essential biosynthetic step is blocked. Cholesta-5,8-dien-3beta-ol, its Delta(5,7) isomer, and other noncholesterol sterols accumulate in subjects with the Smith-Lemli-Opitz syndrome (SLOS), a severe developmental disorder caused by a defe...
